Researchers discover highly potent compound that blocks Ebola virus entry



New compound is more than 100 times more potent than previously reported Ebola entry inhibitors and shows strong protection following oral treatment in a preclinical model

MINNEAPOLIS/ST. PAUL (9/9/2026) — A University of Minnesota-led multi-institutional research team has discovered a highly potent small molecule that blocks Ebola virus entry by stabilizing the virus's entry protein and provides strong protection when given orally in a preclinical model.

The study, recently published in Nature CommunicationsExternal link that opens in the same window, resulted from extensive collaboration among the University of Minnesota, Baylor College of Medicine, Boston University and the University of Florida Scripps Institute, with support from the NIH-funded Midwest Antiviral Drug Discovery (AViDD) Center, based at the University of Minnesota.

MWAC-3634 targets the Ebola virus entry protein. All previously structurally characterized small molecules targeting this protein destabilized it. MWAC-3634 instead stabilizes the entry protein, preventing the shape changes required for viral entry. It is also at least 100 times more potent than previously reported small-molecule Ebola entry inhibitors.

“This compound works in the opposite way from previously studied Ebola entry inhibitors,” said Fang Li, PhD, professor of pharmacology at the University of Minnesota Medical School and co-director of the Midwest AViDD Center. “Earlier compounds destabilized the Ebola entry protein. MWAC-3634 stabilizes it, essentially locking the protein so the virus cannot make the changes required to enter a cell. That reveals a new strategy for blocking infection.”

From billions of molecules to a lead compound

Researchers used DNA-Encoded Chemistry Technology to screen approximately 4.73 billion molecules directly against the Ebola entry protein, uncovering a promising chemical starting point.

“DNA-encoded libraries let us search billions of molecules in a single discovery campaign,” said Srinivas Chamakuri, PhD, assistant director of the Center for Drug Discovery in the Department of Pathology at Baylor College of Medicine. “That scale allowed us to explore chemical space far beyond a conventional screen and uncover a promising starting point we might otherwise have missed.”

Stereochemical analysis of the initial hit identified MWAC-3634 as the most active form. Against authentic Ebola virus, the compound inhibited infection with an IC50 of 0.65 nM and showed no detectable toxicity to cells under the conditions tested.

“The initial hit was only a starting point,” said Peter Dosa, PhD, research associate professor of medicinal chemistry at the University of Minnesota. “The starting compound was a mixture of closely related forms. By identifying the most active form, we substantially increased its potency.”

How the compound locks the entry protein

High-resolution structural studies showed that MWAC-3634 fits into a pocket in the Ebola entry protein and holds it in a stable state. Normally, the protein must change shape for the virus to fuse with and enter a host cell. Locking it in place blocks infection at one of its earliest stages.

“The structure allowed us to see exactly where MWAC-3634 binds and how it stabilizes the Ebola entry protein,” said Dr. Li. “That molecular view helps explain the compound’s high potency and gives us a blueprint for designing improved versions.”

In a preclinical model, oral treatment with MWAC-3634 substantially improved survival, reduced signs of illness and lowered virus levels. The compound also showed 69% oral bioavailability relative to intravenous dosing.

“The protection we saw with oral treatment is particularly encouraging,” said Robert Davey, PhD, professor of virology, immunology and microbiology and interim director of the National Emerging Infectious Diseases Laboratories at Boston University. “This is still an early-stage compound, and additional development and testing will be needed before we know whether it could be useful in people.”

The study combined DNA-encoded library screening, stereochemical analysis, structural biology, authentic Ebola virus testing, drug exposure studies and animal studies across four institutions. It was supported by the National Institute of Allergy and Infectious Diseases, part of the National Institutes of Health, including the Midwest AViDD Center (U19AI171954), and by NIH grant R01AI195592. MWAC-3634 is experimental and has not been approved or shown to prevent or treat Ebola virus disease in humans.

Reports of drug seizures by police rose during first year of decriminalization, SFU study finds​

People in Vancouver experienced an increase in police drug seizures during the first year of British Columbia's drug decriminalization pilot project, according to a Simon Fraser University study.

During the pilot, police in B.C. were not meant to arrest or charge people for possessing drugs for personal consumption, a threshold defined as 2.5 grams. One of the goals of decriminalization was to reduce interaction between the police and people who use drugs, says Kanna Hayashi, co-author of the study. 

“Instead, our study found a significant upward trend of police drug seizures without making arrest,” says Hayashi, professor of health sciences and St. Paul's Hospital Chair in Substance Use Research. 

Although police officially reported Vancouver police officers conducted zero possession seizures below 2.5 grams between February and October 2023, experts say their data—based on self-reports from people who use drugs—allows different parts of the picture to emerge.

Hayashi says the published police-reported statistics only account for events where police officers documented an interaction as being related to drug possession and may or may not have involved an arrest.  

“Our data may have some overlaps with the police-reported data but likely captured events that did not go on to the police records as well. Each dataset is incomplete on its own but offer deeper insights when viewed together,” she says.

The three-year decriminalization pilot ended in January 2026. Published in the journal BMC Public Health, the survey-based study analyzed data from 490 participants in the long running Vancouver Injection Drug Users Study (VIDUS) between March 2022 and February 2024.

Before the decriminalization pilot began Jan. 31, 2023, the quarterly prevalence of police drug seizure without arrest (regardless of the quantity of drugs possessed) were endured by between 0.8 per cent to 2.1 per cent of individuals. During the pilot, that number rose to between 1.1 per cent and 4.1 per cent. 

The study also found participants who reported having drugs seized by police without arrest were three times more likely to experience a non-fatal overdose.

“In a toxic drug-supply crisis, losing a drug supply can have serious consequences,” Hayashi says. “People may feel they need to replace those drugs immediately. If you’re rushing to get new drugs from an unknown source and then rushing to use them to avoid withdrawal symptoms, it may increase the risk of overdose.”

The findings align with concerns long raised by people with lived experience of drug use, says Scotty Archondous, a researcher-member at Police Oversight with Evidence and Research (P.O.W.E.R.).

“As a community-based researcher across multiple projects, I heard directly from many people about their experiences of drug seizures of their personal supply during decriminalization,” says Archondous, who is also a board member of Vancouver Area Network of Drug Users (VANDU).

“We already live so precariously, and these seizures, often without charges or arrest, put our lives at higher risk of violence and harm based on what comes next: debts, desperation and sometimes disrupted access to a trusted source.” 

For Hayashi, data informed by people with lived experience must be viewed in tandem with official police statistics.

“Even though the pilot has ended, it's important to understand what happened and what can be improved. Those lessons can help shape future decriminalization policies here and elsewhere,” she says.

 

Body-related TikTok content linked to self-objectification and mood, study shows

Viewing “thinspiration” content on TikTok increases self-objectification and worsens mood, while viewing body-positive content also increases self-objectification but is linked to more positive mood, according to a new study published September 9, 2026 in the open access journal PLOS One by Samantha Jones of Western University, Canada, and colleagues.

Self-objectification occurs when people feel their value is determined by their appearance, viewing themselves and their bodies as objects. Appearance-focused social media content includes thinspiration, which depicts a thin-ideal and promotes weight loss; fitspiration, containing fat-loss promotion messages; and body positivity content, promoting body acceptance and challenging thin/fit ideals. Recently, researchers have questioned the beneficial nature of body positivity content, as well as the overall associations between self-objectification and mood.

In the new study, researchers recruited 323 women and feminine-identifying people who reported their general disposition to self-objectify before watching a randomized set of 20 TikTok videos spanning five categories: thinspiration, fitspiration, body positivity, general positive, and neutral. After each video, participants reported their self-objectification and mood levels.

Compared with watching positive videos, self-objectification was highest after viewing thinspiration (B=1.24), followed by body positive (B=0.98) and fitspiration (B=0.43) content. Thinspiration and fitspiration were both associated with increased negative mood and decreased positive mood compared with neutral videos. Body positive content, on the other hand, was associated with increased positive mood. Greater self-objectification after viewing body positive content strengthened this positive mood effect (B=6.49).

The study measured only momentary effects of viewing videos, so cannot determine longer-term effects, and participants did not complete a baseline mood or self-objectification measure before viewing videos. The sample was young and consisted largely of women of average body size, limiting generalizability.

However, the researchers conclude that appearance-focused social media content broadly increases self-objectification, but this is not always associated with negative outcomes. Future research could probe into how to lessen the impacts of thinspiration and fitspiration on mood, they say, especially for those with a high tendency to self-objectify.

The authors add: “We found that women experienced negative emotions when viewing social media videos that idealize thinness. Women who tend to value their appearance over their other qualities were especially vulnerable to negative mood after viewing thin-ideal videos. However, in the moment, these same appearance-focused thoughts may serve to boost mood when women engage with body positive videos.” 

“Presenting videos to participants in a randomized order and investigating each video’s impact on mood allowed us to disentangle the effect that different forms of content have on women’s mood while viewing videos from TikTok.” 

“Unpredictable exposure to a variety of TikTok videos can elicit shifts in both thinking and feeling in the moment.”

Over three-thousand scientific article titles across disciplines have made use of Beatles song titles and lyrics, according to a study published September 9, 2026, in the open access journal PLOS One by Gabriel Budel of Delft University of Technology in The Netherlands, and colleagues. This analysis is an unprecedentedly large-scale, cross-disciplinary account of how the Beatles have left their mark on scientific writing.

Researchers routinely draw on cultural touchstones to frame their work, and popular music in particular has left a distinct mark on academic publishing. Of all musical artists to have influenced scientific communication in this way, the Beatles may be one of the most pervasive.

In the new study, Budel and colleagues examined how often Beatles song titles and lyrics appear in academic article titles. The researchers identified 2,237 exact references and 963 instances of creative wordplay in data from the academic database Scopus. Using a curated list of Beatles songs and lyrics, they combined automated title queries with state-of-the-art large language models to detect both exact references and wordplay across disciplines. The analysis revealed that the references are not confined to any single field. Medicine and the Social Sciences dominate, but the long tail reaches fields as varied as Computer Science, Economics, Environmental Science, and the Arts and Humanities.

In addition, the top ten Beatles song titles appear in 1,584 article titles, more than the equivalent top tens of Michael Jackson (65 articles) and Elvis Presley (196 articles), the two next best-selling artists. The most frequently referenced song titles—‘The Long and Winding Road’ (798 hits), ‘With a Little Help from My Friends’ (209 hits), and ‘Here, There and Everywhere’ (129 hits)—are apt metaphors for the scientific process, including the course of research, collaboration, and discovery. Many variations on ‘All You Need Is Love’ made their way into the academic literature, such as ‘All you need is sleep.’ The 2017 article ‘Attention is all you need’ is one of the most influential scientific publications of the 21st century to date. It was cited 89,737 times on Scopus.

As noted by the researchers, there is a clear practical lesson for authors: a well-chosen cultural reference can make a technical contribution more memorable, as long as the allusion fits the content without sacrificing clarity for readers or discoverability in literature searches. The same titles can help editors and science communicators reach past a specialist audience, since a shared cultural touchstone travels easily between fields. Overall, the study advances the understanding of how popular culture shapes academic communication.

Gabriel Budel adds: “We found more than 3,000 scientific papers whose titles borrow from the Beatles, and the most-cited one is 'Attention is all you need,' the 2017 paper that invented the architecture behind ChatGPT. Its title is a play on 'Love is all you need.' We then used exactly that technology to find the rest of the Beatles wordplay hiding in the literature.”

“Some of these are genuinely brilliant. 'The lung and winding road' is a paper on Long COVID that changes one vowel. 'Here comes the SU(N)' turns 'Here Comes the Sun' into a quantum computing paper about mathematical groups. There's a patent law article called 'Everybody's got something to hide except me and my patented monkey.' Someone was having a very good day at their desk.”

“The funniest part of the job was also the most tedious. The language model flagged thousands of candidate titles, and the three of us then read every single flagged title by hand to decide whether it was really a Beatles pun. We sat there arguing about whether 'You say you want a resolution' counts. It does.”

“There's a nice irony in the timing. We used a small language model to hunt for Beatles wordplay, and its weak point was attribution: it knew a pun when it saw one, but couldn't reliably tell you which song it came from. Newer models handle that easily. The models born out of 'Attention is all you need' learned to hear 'Love is all you need' faster than we could finish the paper.”

Michiel Tjepkema adds: “I teach law and I co-host a Beatles podcast, so I've spent years arguing that the band's reach goes far beyond music. This is the first time I've been able to put a number on it. What strikes me most is that scientists tend to invert the Beatles' optimism: 'With a little help from my enemies,' 'Although it helps, love is not all you need.' The songs are hopeful; the papers are skeptical.”

Robert E. Kooij adds: “As network scientists we're used to seeing power laws in earthquakes, in social networks, in wealth. It turns out references to Beatles songs follow one too: a handful of songs are cited constantly and the bulk of them are used once or twice. The Beatles apparently obey the same statistics as the rest of nature.”
